摘要: Abstract Different classes of chemicals can induce a phototoxic effect by absorbing light energy within the wavelength range sunlight. The assessment photo-safety is therefore an obligatory part development new drugs. Ten UV–vis (280–800 nm)-absorbing compounds (ketoprofen, promazine, chlorpromazine, dacarbazine, acridine, lomefloxacin, 8-methoxypsoralen, chlorhexidine, titanium dioxide, octylmethoxycinnamate) were tested for their photogenotoxic potential in alkaline comet assay presence and absence UV–vis. In order to establish easy timesaving protocol photo screening test, application 96-well plates was essential. use mouse lymphoma L5178Y cells, cell line growing suspension, allowed determination photocytotoxicity with Alamar Blue photogenotoxicity parallel. cells incubated test 20 min irradiated simulated sunlight from 280 800 nm. applied UV dose 600 mJ/cm2 UV-A 30 mJ/cm2 UV-B. After post-incubation 10 min, performed. All which are known be (8-methoxypsoralen, ketoprofen, lomefloxacin) showed positive under our conditions. Furthermore, four not (promazine, analysed. For none them increase DNA damage following irradiation observed this study. conclusion, all chemical classified agreement published data. From data presented it concluded that reliable model assess photochemical genotoxicity vitro.
