DNA–RNA hybrid formation mediates RNAi‐directed heterochromatin formation

摘要: Certain noncoding RNAs (ncRNAs) implicated in the regulation of chromatin structure associate with chromatin. During formation RNAi-directed heterochromatin fission yeast, ncRNAs transcribed from are thought to recruit RNAi machinery for heterochromatin; however, molecular details this association not clear. Here, using RNA immunoprecipitation assay, we showed that heterochromatic ncRNA was associated via a DNA-RNA hybrid and bound RNA-induced transcriptional silencing (RITS) complex. The presence cell also confirmed by immunofluorescence analysis anti-DNA-RNA antibody. Over-expression depletion RNase H vivo decreased increased amount formed, respectively, both disturbed heterochromatin. Moreover, formed on, over-expression inhibited of, artificial induced tethering RITS mRNA. These results indicate retained on hybrids provide platform assembly suggest plays role chromatic function.
