摘要: Background: Differentiation of mesenchymal stem cells (MSCs) to hepatocyte-li ke could be associated with development liver function factors. The impact differentiation-dependent changes on DNA integrity is not well understood. In this study, hepatocytes and their progenitor were treated aflatoxin B1 (AFB1) amplification selected genes linked damage was examined. Methods: MSCs CD34 + isolated from umbilical cord blood (UCB) AFB1 (0, 2.5, 10 20 µM) in selective media supporting the hepatocyte differentiation. After 24 htreatment (Comet assay) rates P53 -globin measured using real time polymerase chain reaction (QPCR). Results: results show that treatments resulted a concentration- dependent increase suppression specific gene amplification. extent significantly greater differentiated when compared those obtained cells. effects rate QPCR showed lesions (expressed as lesions/10 kb) derived Conclusions: These data together cytochrome P450 (CYP3A4) expression suggest non-differentiated are probably less vulnerable genotoxic agents them.
