Flavokawain B inhibits the growth of acute lymphoblastic leukemia cells via p53 and caspase-dependent mechanisms

摘要: The development of novel chemotherapeutic drugs is needed for the treatment patients with acute lymphoblastic leukemia (ALL). In this study, anti-leukemic effect and potential molecular mechanisms action flavokawain B on ALL were investigated. Flavokawain was found to significantly inhibit cellular proliferation B-ALL T-ALL cell lines in a dose-dependent manner. It also induced apoptosis by increasing expression p53, Bax Puma, activating cleavage caspase-3 poly ADP-ribose polymerase (PARP). Furthermore, enhancement p53-dependent could be rescued pifithrin-α, pharmacological inhibitor p53 transcriptional activity. Moreover, blast cells from 16 inhibited B, tumor growth xenograft mice suppressed drug. conclusion, our results demonstrate therapeutic ALL.
