Application of a novel method of double APAAP staining with subsequent quantitative image analysis to the examination of integrin expression in undifferentiated-type gastric carcinomas.

摘要: In undifferentiated-type gastric carcinoma (UGC), recognition of cancer cells is not easy, which has hampered its precise phenotypic analysis. To examine alterations the integrin phenotype during progression UGC, we used double alkaline phosphatase anti-alkaline staining and computer-aided image analyses for expression α1, α2, α3, α5, α6, αV, β1, β4 subunits αVβ3, αVβ5, αVβ6 integrins in cytokeratin-positive mucosal, submucosal, deeper parts 10 early 17 advanced UGCs, their non-neoplastic counterparts, 9 lymph node (LN) metastases. We revealed declining epithelial (α2, β4) increasing mesenchymal (α1, α5) as tumor invaded deeper, reflecting gradual epithelial-to-mesenchymal transition invasion. Enhanced αV subunit αVβ3 αVβ5 correlated with invasion, that LN metastasis. Our results have demonstrated method introduced suitable analysis dynamic repertoire UGC progression.
