Novel type of general protein assay using a chromogenic and fluorogenic amine-reactive probe.

摘要: We report on a new and simple one-reagent method for general protein assay. It makes use of one two reactive labeling reagents presented here (and referred to as pyrylium [Py] labels). These can be applied both photometric fluorometric assays at near neutral pHs room temperature. The Py labels undergo large spectral change conjugation the amino group proteins typically their color from blue red. Therefore, unlike in other assays, there is no need separate unconjugated (blue) label red conjugate, which determined by direct photometry with limit detection 1.2 μg/ml human serum albumin. assay extended fluorometry because fluorescence free weak (with quantum yield 40%) conjugation. strong conjugates directly without interference weakly fluorescent) label. resulted 60 ng/ml bovine albumin (BSA). Validation blood plasma samples spiked BSA gave recoveries range 91 103%.
