摘要: Previous data suggest that nucleotides are important mitogens in the developing retina. Here, effect of ATP on death cultured chick embryo retina cells was investigated. In cultures obtained from retinas 7-day-old embryos (E7) were cultivated for 2 days (E7C2), both and BzATP induced a ∼30 % decrease cell viability time- dose-dependent could be blocked by 0.2 mM oxidized or 0.3 μM KN-62. An increase cleaved caspase-3 levels number TUNEL-positive observed when incubated with 3 immunolabeling cleaved-caspase over neurons but not glial cells. ATP-dependent developmentally regulated, maximal being detected E7C2-3. Nucleotides able to neuronal ethidium bromide sulforhodamine B uptake mixed purified cultures, an antagonists Brilliant Blue G ATP. contrast, nucleotide-induced only glia. ATP-induced glutamatergic MK801 DNQX activation P2X7 receptors decreased [3H]-d-aspartate concomitant accumulation it extracellular medium. These results induces apoptosis retinal culture through glutamate ionotropic receptors. Involvement receptor-mediated inhibition is suggested.
