Description of a multiplex Bordetella pertussis and Bordetella parapertussis LightCycler® PCR assay with inhibition control

摘要: While culture for Bordetella species is highly specific, sensitivity extremely variable due to patient age, immunization status, antibiotic treatment, and specimen transport conditions. We evaluated a real-time multiplex PCR assay as an alternative the detection differentiation of pertussis parapertussis. The conditions allowed simultaneous one B. organism five parapertussis organisms per reaction. An inhibition control was incorporated into assay. Of 163 total samples evaluated, 37 38 positive by either or direct fluorescent antibody testing (DFA) were also (97% sensitivity). 125 culture- DFA-negative samples, 101 negative (81% specificity). described rapid, sensitive, contamination-limiting, that controls inhibition. performs well using liquid swab from dried material on slides.
