摘要: Telomerase is a ribonucleoprotein complex with reverse-transcriptase activity responsible for telomere reconstitution. High telomerase was found in cancer cells, but not differentiated homologous nonmalignant tissues. We demonstrated previously that the disappearance of reliable marker tumor cell killing human lines. have investigated possibility evaluating chemosensitivity neoplastic cells different origin [ovary, lung, breast, gastrointestinal, skin (melanoma)] obtained from patients, by measuring residual after drug treatment vitro. Using classical telomeric repeat amplification protocol ("TRAP") assay based on polymerase chain reaction, we examined untreated or drug-treated suspensions, derived processing surgical specimens. Feasibility and reproducibility were evaluated according to various parameters, including concentration, time vitro culture, type tumor. The results indicated highly sensitive reproducible, can be performed using specimens reasonable percentage cases, ranging 40% (breast cancer) 100% (ovarian cancer). Moreover, provides comparable wide range presence normal does interfere results. Prolonged culture required because completed within 24 72 hours sample collection. In conclusion, present investigation technical bases future studies evaluate whether this would able predict patient's response antitumor agents.
