Development of specific PCR assays for the detection of Cryptocaryon irritans

摘要: Cryptocaryon irritans is one of the most important protozoan pathogens marine fish, causing “white spot” disease and posing a significant problem to aquaculture. In present study, C. irritans-specific reverse primer (S15) was designed based on published sequence second internal transcribed spacer (ITS-2) ribosomal DNA (rDNA) used together with conserved forward P1 develop specific polymerase chain reaction (PCR) assay for direct, rapid, detection irritans. The specificity these primers tested both closely distantly related ciliates (Pseudokeroronpsis rubra, Pseudokeroronpsis carnae, Euplotes sp. 1, Ichthyophthirius multifiliis, Pseudourostyla cristata, Paramecium caudaium), only detected no product amplified from any other examined in this study using set P1-S15. PCR able detect as low 45 pg nested two sets (P1/NC2, P1/S15) increased sensitivity, allowing single species-specific assays should provide useful tools diagnosis, prevention, molecular epidemiological investigations infection fish.
