Regulation of voltage-gated sodium channel expression in cancer: hormones, growth factors and auto-regulation.

摘要: Although ion channels are increasingly being discovered in cancer cells vitro and vivo, shown to contribute different aspects stages of the process, much less is known about mechanisms controlling their expression. Here, we focus on voltage-gated Na(+) (VGSCs) which upregulated many types carcinomas where activity potentiates cell behaviours integral metastatic cascade. Regulation VGSCs occurs at a hierarchy levels from transcription post-translation. Importantly, mainstream mechanisms, especially hormones growth factors, play significant role regulation. On whole, major hormone-sensitive cancers, such as breast prostate cancer, there negative association between genomic steroid hormone sensitivity functional VGSC Activity-dependent regulation by positive feedback has been demonstrated strongly whereby self-sustaining, with its promoting further channel Such auto-regulation unlike normal activity-dependent mostly via feedback. Throughout, highlight possible clinical implications expression cancer.
