摘要: Digital ELISA is introduced as a novel platform with unique advantages for detecting multiple kinds of single-molecule in the sample. How to improve sensitivity detection direction current related research. Here, we report an immunoassay method that applied electrokinetic effects isolate individual encoded beads and confine micro-wells efficiency cytokines simultaneously. The microfluidic design provided non-uniform electric field induce dielectrophoresis (DEP) force manipulate beads. Finally, concentration captured obtained by picking up each bead from image then integrating intensity fluorescent emitted results demonstrated fill percentage raised 10-20% 60-80% DEP effect. By comparing fluorescence color particle, itself its surface, four target cytokines, IL-2, IL-6, IL-10 TNF-α, were calculated pg/ml level. spike recovery experiments verified efficiency, more than 70% molecules captured. reliability our flow cytometry well. In conclusion, expect application can increase digital rapid detection.
