摘要: Abstract l -asparaginase is an enzyme of medical prominence and reputable as a chemotherapeutic agent. It also has immense potential to cure autoimmune infectious diseases. The vast application this in healthcare sector increases its market demand. However, presently the huge demand not achieved completely. This serves basis explore better producer microbial strains bridge gap between supply therapeutic enzyme. present study deals with successful screening potent microorganisms producing -asparaginase. 47 were screened including bacteria, fungi, yeasts. Among all, Penicillium lilacinum showed highest activity i.e., 39.67 IU/ml. Shigella flexneri 23.21 IU/ml (highest among all bacterial strain tested). Further, 3-D structure from higher was developed validated silico for activity. -asparagine (substrate -asparaginase) docked inside binding pocket P. S. flexneri. Docking score most common substrate −6.188 (P. lilacinum), −5.576 (S. flexneri) which quite good. Moreover, chemical property revealed that amino acid residues Phe 243, Gln 260, Gly 365, Asp 386 181, Thr 318, Asn 320 have important role H-bonding. results supports strengthen wet lab results. outcome obtained motivates take result industry economic/massive production diverse application.
