摘要: Methods for 3D histology reconstruction from sparse 2D digital images depend on knowledge about the positions, orientations, and deformations of tissue slices due to the process. This work quantitatively evaluates typical assumptions position orientation whole-mount prostate sections within coarsely sliced tissue blocks deformation during histological processing sectioning. 3-5 midgland tissue blocks each 7 radical prostatectomy specimens were imaged using magnetic resonance imaging before histology processing. After standard paraffin sectioning, resulting digitised. Homologous anatomic landmarks identified 22 MR images. Orientations depths of sections relative front faces measured based best-fit plane through landmarks on The mean±std section was 1.7±1.1° depth 1.0±0.5 mm. Deformation assessed by four transformation models (rigid, rigid+scale, affine thin-plate-spline (TPS)) align images, evaluating measuring target registration error (TRE) a leave-one-out cross-validation. rigid model had higher mean TRE (p<0.001) than the other models, rigid+scale TPS (p<0.001 p<0.01 respectively). These results informed design development method reconstruction based extrinsic strand-shaped fiducial markers which yielded 0.7±0.4 mm TRE.
