Selective detection of N6-methyladenine in DNA via metal ion-mediated replication and rolling circle amplification

摘要: N6-methyladenine (6mA) is reported as a potential epigenetic marker in eukaryotic genomes. However, accurate identification of the location 6mA DNA remains challenging task. Here, we show that Ag+ can selectively stabilize A–C mismatch and efficiently promote primer extension. In contrast, complex 6mA–Ag+–C instable therefore cannot be recognized by polymerases, resulting termination Based on this finding, successfully identified quantified at single-base level through analysis gel bands extended primers fluorescence measurements combined with rolling circle amplification. The high selectivity sensitivity strategy may provide new platform for efficient future.
