摘要: Low cell competency for regeneration and transformation is the main cause of so-called recalcitrance to transform a species or genotype. A research was conducted determine optimum conditions in vitro plant involving organogenesis Citrus aurantium, which an important rootstock worldwide. Seeds with peeled teguments were germinated vitro, either kept dark 6 weeks maintained absolute 4 followed by 10 days 16-h photoperiod (56 mumol m-2 s-1) at 27 +/- 2 degreesC. Epicotyl-originated explants cultured MS medium supplemented 6-benzylaminopurine (BAP) (0, 1, 3 mg L-1) Naphthaleneacetic acid (NAA) 0.05, 0.1 0.2 induce organogenesis. Effects Pre-culture liquid 1 days) on number responsive (RE) have been also evaluated. In next step, having buds transferred containing Gibberellic Acid (GA3) 0.5 size shoots, produced RE are then measured. The highest percentage (90%) obtained using 2.5 L-1 BAP combination 0.05 NAA had pre-culture period epicotyls allowing grow darkness weeks, s-1). well-developed shoots 4.2 per explant GA3. These protocols suitable association aurantium Agrobacterium-mediated genetic transformation.
