Isolation of the minor and rare constituents from fruits of Peucedanum alsaticum L. using high-performance counter-current chromatography.

摘要: A high-performance counter-current chromatography (HPCCC) method was applied for the first time preparative separation and purification of three rare compounds which occur as minor constituents in fruits Peucedanum alsaticum L.: 5-substituted coumarin notoptol two dihydropyranochromones: divaricatol ledebouriellol. scale-up process from analytical to a very short developed. In order purify range with different polarity separate experiments were performed, one reverse phase, other normal using same crude extract. two-phase solvent system composed n-hexane/ethyl acetate/methanol/water (1:1:1:1) The components purified collected analyzed by liquid chromatography. yielded 0.7 mg notoptol, 1.46 ledebouriellol at purity 99.5%, 10 mixtures divaricatol, alsaticol alsaticocoumarin, where present 22% peak area. These amounts obtained 1 g extract single run. This is when ledebouriellol, isolated run HPCCC these identified plant genus.
