Interactomic approach for evaluating nucleophosmin-binding proteins as biomarkers for Ewing's sarcoma.

摘要: Nucleophosmin (NPM) is a novel prognostic biomarker for Ewing's sarcoma. To evaluate the utility of NPM, we conducted an interactomic approach to characterize NPM protein complex in sarcoma cells. A gene suppression assay revealed that promoted cell proliferation and invasive properties FLAG-tag-based affinity purification coupled with liquid chromatography-tandem mass spectrometry identified 106 proteins complex. The functional classification suggested participates critical biological events, including ribosome biogenesis, regulation transcription translation, folding, are mediated by these proteins. In addition JAK1, candidate sarcoma, complex, includes 11 known as biomarkers other malignancies. Meta-analysis expression profiles 32 patients 6 were significantly independently associated survival period. These observations suggest role well value Further, our study suggests potential applications interactomics conjunction meta-analysis discovery.
