摘要: Abstract A simplification of the enzymatic isotopic assay for serotonin is described, Serotonin converted to [ 3 H]melatonin by a two-step reaction: N -acetylation using acetic anhydride, followed O -methylation with enzyme hydroxyindole -methyltransferase (EC 2.1,1.4) and S -adenosyl- l -[ methyl - H]methionine as donor. The present avoids use unstable acetylating enzyme, rat liver -acetyltransferase (EC2.3,1.5). Blank values are lowered considerably sensitivity doubly increased. Two-tenths micromole per 30 μl sample in tissue homogenates can be measured.
