Application of thin-layer chromatography image analysis technique in quantitative determination of sphingomyelin

摘要: A sensitive and convenient method for sphingomyelin determination was developed based on thinlayer chromatography (TLC) image-processing analysis. The mobile phase composition, detection quantification conditions were systematically investigated through several trials. molybdenum blue reaction allowed specific of the phospholipid with a high sensitivity wide linear range. Digital images TLC plate chromatograms captured flatbed scanner converted into peak using TLSee® software quantitative analysis conducted. linearity, sensitivity, accuracy precision system evaluated. limits 0.5 1.7 μg/spot, respectively. Separation mixture consisting sphingomyelin, phosphatidylcholine, phosphatidylethanolamine also carried out.
