Pulmonary TCR γδ T cells induce the early inflammation of granuloma formation by a glycolipid trehalose 6,6'-dimycolate (TDM) isolated from Mycobacterium tuberculosis.

摘要: We previously showed that formation of pulmonary granulomas in mice response to a mycobacterial glycolipid, trehalose 6,6'-dimycolate (TDM) is due the action TNF-α and not IFN-γ. However, mechanisms maintenance are yet clear. The purpose present study evaluate granuloma by TDM at early phase. Histological analysis inflammatory cells infiltrated murine interstitium on day 2 after an intravenous injection with as w/o/w emulsion. Clear was observed 7 injection. mRNA expression IL-17, IFN-γ macrophage protein found lung mononuclear major IL-17-producing were T-cell receptor (TCR) γδ T expressing Vγ6. In depleted treatment anti-TCR monoclonal antibody, number TDM-induced decreased, but size affected. Our results suggest glycolipid causes activation TCR stimulates chemotaxis including neutrophils lung.
