摘要: Intervertebral disk degeneration is a progressive and debilitating disease with multifactorial causes. Nitric oxide (NO) might contribute to the cell death pathway. We evaluated presence of constitutive form neuronal NOS (nNOS) in both health degenerated intervertebral through qPCR immunohistochemistry. also analyzed potential role nNOS modulation tail needle puncture model degeneration. Male Wistar rats were submitted percutaneous 21-gauge coccygeal vertebras. The selective pharmacological inhibitor N (ω)-propyl-l-arginine (NPLA) or nNOS-target siRNA (siRNAnNOShum_4400) was injected immediately after 30-gauge needle. Signs by vivo magnetic resonance imaging histological score. found that intact disks express low levels mRNA. Disk injury caused 4 fold increase mRNA content at 5 h post lesion. However, NPLA slight mitigate degenerative progress. Our data show evidence its upregulation during Further studies would disclose therapeutical value
