摘要: Several plasmids with different promoter sequences, all containing the HygB gene that confers resistance to antibiotic hygromycin B, were evaluated for their ability transform Trichoderma viride strain T 105-227 Nic-. Plasmid pH IB, which contains a sequence from Cochliobolus heterostrophus, was found give largest number of putative transformants. Transformation T. harzianum strains T12-2 His- and T95-1 Lys- attempted, time at transformed protoplasts exposed B be critical. If added before had regenerated least 6 h, no transformants obtained. However, if regeneration proceeded more than 12 many nontransformed formed thalli. Putative heterokaryotic. Even thalli grown in presence nuclei low; only 0.5 0.8% conidia capable producing B. Subcultures derived single B-resistant stable. Southern analysis indicated plasmid integrated into genome either tandem repeat pattern or copy insertion.
