摘要: · Purpose: It has been previously shown that CD4+ T-lymphocytes are critical mediators in HSV-1 stromal keratitis (HSK). T cell subpopulations (type 1, type 2) can be defined by their capabilities of producing different sets cytokines. This study was performed to determine the role 1 and 2 cytokines murine HSK. Methods: BALB/c mice (n=20) were inoculated with 105 PFU (KOS strain) followed clinically. At various time points post-infection (p.i.), conjunctival corneal tissues analyzed histologically (n=2 each point), immunohistochemically (n=5 point) for presence interleukin-1α (IL-1α), (IL-2, interferon-γ) a cytokine (IL-4). The expression mRNA tested eye samples reverse transcription-polymerase chain reaction (RT-PCR). Results: Stromal clinically progressed after day 9. In 15% mice, disease regressed until 14 p.i. Polymorphonuclear neutrophils, lymphocytes other mononuclear cells infiltrated conjunctiva rapidly expanded central cornea between days 7 14. IL-1α, IFN-γ IL-2 found eyes at IL-1α protein detected conjunctiva, limbus epithelium 2. staining intensities increased progression. paralleled intensities. contrast, IL-4 through infection; compared IFN-γ, lower. Conclusions. findings suggest lymphocytic infiltrate during development is predominantly composed expressing IFN-γ. Type participate late stage inflammation might useful improve course disease.
