Phenotypic drug resistance in mammalian cells in vitro.

摘要: When mammalian cells are cultured at low concentrations of toxic drugs, they often become phenotypically resistant. We studied whether this phenotypic resistance is due to selection preexisting variants. The drugs 8-azaguaine (AG) and 6-thioguanine (TG) were used and, as a parameter for resistance, the incorporation hypoxanthine was determined. Preexisting variation among clones in uptake found, has hereditary component. This transmission aberrant does not appear stable trait, cell lines returned gradually original steady state. There indications that within population with altered levels arise continuously high frequency. Treatment marginally AG or TG indicates that, least AG, survival related uptake. observed phenomena could be importance cancer chemotherapy.
