摘要: Inactivation of a chromosomal gene is useful approach to study the function in question and can be used produce desired phenotype organism. This chapter describes how generate such mutants cyanobacterium Synechococcus sp. PCC 7002 green sulfur bacterium Chlorobium tepidum by natural transformation with synthetic DNA constructs. Two alternative methods constructs, both performed entirely vitro based on polymerase chain reaction (PCR), are also presented. These ligation fragments T4 ligase, megaprimer PCR.
