摘要: Assisted reproductive technology (ART) is being increasingly applied to overcome infertility. However, the in vitro production process, main procedure of ART, can lead aberrant embryonic development and health-related problems offspring. Understanding mechanisms underlying ART-induced side effects important improve ART process. In this study, we carried out comparative transcriptome profiling between vivo- (IVO) vitro- produced (IVP) mouse blastocysts. Our results suggested that actin organization might be a major factor contributing impaired IVP embryos. To test this, examined effect disorganization on preimplantation Specific disruption by cytochalasin B (CB) indicated well-organized essential for development. Supplementing culture medium with 10–9 M melatonin, cytoskeletal modulator adult somatic cells, significantly reversed disrupted expression patterns genes related organization, including Arhgef2, Bcl2, Coro2b, Flnc, Palld. Immunofluorescence analysis showed melatonin treatment embryos improved distribution filaments (F-actin) from 8-cell stage onwards. More importantly, found alleviated CB-mediated F-actin rescued CB-induced This first study indicate implicated during stage.
