Cell proliferation after ischemic injury in gerbil brain. An immunocytochemical and autoradiographic study.

摘要: Tritiated thymidine autoradiography was used to measure cellular proliferation after ischemic injury in gerbil brain. Gerbils were subjected bilateral occlusion of the common carotid arteries which resulted areas necrosis, or infarcts, posterior thalamus midbrain. From 12 h 10 days following ischemia, gerbils injected with 3H thymidine, sacrificed 4 later, and brains sectioned. In order identify astrocytes monocytes/macrophages, immunocytochemistry performed prior autoradiography, using antisera against glial fibrillary acidic protein endothelial-monocyte reticuloendothelial antigen, respectively. Immunocytochemistry also visualize microvessel laminin, myelin, leakage serum albumin. Lastly, a histochemical procedure for acid phosphatase activity employed verify phagocytic wound. A reproducible sequence reactions took place during first ischemia. Early changes included albumin myelin breakdown, followed by arrival monocytes at 2 their differentiation into macrophages 5 days. These cells exhibited intense from 6 post-ischemia. Microvessel endothelial maximally labeled Hypertrophied apparent proliferated 3 7 post-ischemia, wound replaced “glial scar”.
