Improvement of recombinant endoglucanase produced in Pichia pastoris KM71 through the use of synthetic medium for inoculum and pH control of proteolysis

摘要: The long lag time in basal salts medium (BSM) and an occurrence of proteolysis are major problems for recombinant protein production Pichia pastoris KM71. In this study, optimal conditions were explored fed-batch cultivation fungal endoglucanase P. pastoris It was found that process times much reduced when the synthetic FM22 used inoculum compared with enriched buffered glycerol complex (BMGY) medium. highest activity obtained at 30°C which more than 10 fold higher produced from shake flask. At 30°C, specific dependent on culture pH a observed 5.0 6.0. likely due to lower rate proteolysis, since truncated species apparent 6.0, but not 5.0. Thus, is condition suitable economical large scale. combination using control by growth could be applied other proteins P. pastoris.
