Enhancement of thermostable β-glucosidase production in a slow methanol utilization strain of Pichia pastoris by optimization of the specific methanol supply rate

摘要: Control of the methanol supply during production stage is a crucial parameter for maintaining cell growth and enhancing recombinant protein by Pichia pastoris. In this study, optimization initial specific rate was explored high density fed-batch cultivation fungal β- glucosidase in slow utilization strain (MutS), P. pastoris KM71. By varying feed rates initiating induction at concentration 60 g/L, optimum determined to be 30.34 ± 0.34 mg/g·h. A proposed according applied β-glucosidase higher concentrations 80 100 g/L. The highest activity obtained 2851.7 14.6 U/mL, which four times than that with reference condition (40 g/L concentration). success approach suggests strategy optimizing could adopted other proteins employing inducible system.
