Oxygen-limited fed-batch process: an alternative control for Pichia pastoris recombinant protein processes

摘要: An oxygen-limited fed-batch technique (OLFB) was compared to traditional methanol-limited (MLFB) for the production of recombinant Thai Rosewood β-glucosidase with Pichia pastoris. The degree energy limitation, expressed as relative rate respiration (qO/qO,max), kept similar in both types processes. Due higher driving force oxygen transfer OLFB, and methanol consumption rates were about 40% OLFB. obligate aerobe P. pastoris responded severe limitation mainly by increased maintenance demand, measured carbon dioxide per methanol, but still somewhat cell density (5%) product concentrations (16%) obtained. viability similar, 90–95%, process types, amount total proteins released medium much less OLFB processes resulting substantially (64%) specific enzyme purity input downstream processing.
