摘要: The α-glucosidase (AGL) from Aspergillus niger has been applied to produce isomaltooligosaccharides. In the present study, various factors which affect yield of recombinant AGL, produced by engineered Pichia pastoris, were investigated. expression level reached 5.5 U ml−1 in bioreactor after optimization parameters initial induction cell density, temperature and methanol concentration. addition, it was found that coexpression protein disulfide isomerase (PDI) inhibited growth P. pastoris strains had an adverse effect on production while codon native A. encoding gene (aglu) resulted a significant enhancement enzyme production, 10.1 U ml−1. We believe AGL is increased as result enhanced translation efficiency well more stable mRNA secondary structure. contrast, PDI under control alcohol oxidase promoter (PAOX1) seems be less efficient helping bond formation probably induce unfolded response, further leads apoptosis degradation.
