摘要: Granzyme B (GrB) is the primary molecular mediator of apoptosis by cytotoxic T lymphocytes (CTLs) and natural killer (NK) cells. It a unique mammalian aspartic acid-cleaving serine protease. On cell receptor activation, GrB released from CTL cytoplasmic granules exocytosis, enters target cells and, in presence granule pore-forming protein perforin, it initiates processing caspases apoptosis. also activated adenovirus, which can effectively replace perforin. Methods for purification quantitation preparation titration are described. In addition, methods application these reagents to initiation tumor cells, with several assays detecting apoptotic activity, detailed.
