摘要: In vivo electroporation is a powerful technique for the introduction of genes into organisms. Temporal and spatial regulation expression introduced genes, or RNAi, would further enhance utility this method. Here we demonstrate conditional gene from electroporated plasmids in postnatal rat retina embryonic mouse brain. For temporal regulation, Cre/loxP-mediated inducible vectors were used combination with vector expressing conditionally active form Cre recombinase, which activated by 4-hydroxytamoxifen. Onset was regulated timing 4-hydroxytamoxifen administration. transgenes expressed using promoters specific rod photoreceptors, bipolar cells, amacrine Muller glia progenitor cells. Combinations these constructs will facilitate variety experiments, including cell-type-specific misexpression, fate mapping precursor
