Spread of clonal T-cell expansions in rheumatoid arthritis patients.

摘要: Abstract Despite a large number of studies identifying expanded T-cell clones among infiltrating lymphocytes, little is known about their distribution in patients suffering from rheumatoid arthritis. To evaluate the clonality α/β populations arthritic locations and PBL, we determined CDR3 size lengths TCR β-chain transcripts using BV (Vβ), BC (Cβ), BJ (Jβ), clonotype-specific primers. Transcripts PBL healthy donors show gaussian profiles approximately eight peaks most subfamilies. Dominant standing out above normal background identify expansions one or several within given subfamily. The analysis six arthritis showed clonal all samples including PBL. Synovial tissue infiltrates revealed less complex repertoires with greater than Expanded varied patient to another; no recurrences were observed. Most interestingly, identical identified bilaterally knee joints same patient. Our data that are not only locally but can also be found periphery, strongly suggest many similar spread throughout bodies patients.
