Muco-adhesive clarithromycin-loaded nanostructured lipid carriers for ocular delivery: Formulation, characterization, cytotoxicity and stability

摘要: Abstract Topical ophthalmic formulations are the preferred approach to treat anterior segment of eye as it is a non-invasive therapeutic approach. The ocular bioavailability drugs generally limited, due presence impervious anatomical barriers and low residence time contact with target tissue. Optimization clarithromycin-loaded nanostructured lipid carriers using Design Experiments was undertaken. Manufacture achieved hot emulsification ultrasonication. Formulation process parameters were successfully identified following screening subsequently optimized Tween® 20, stabilizer. Muco-adhesive properties that could potentially increase time, in vitro clarithromycin release cytotoxicity against HeLa cells evaluated. Short term stability studies lipidic assessed at 4 °C 22 °C. formulation exhibited muco-adhesive under stationary conditions Laser Doppler Anemometry, sustained API over 24 h conditions. In revealed NLC less cytotoxic comparison pure API. results suggest may have potential enhance precorneal retention, availability permit dose reduction or use longer dosing frequency.
