摘要: Immune interferon (IFN-gamma), endogenously labeled with [35S]methionine, was produced in human peripheral blood lymphocyte cultures stimulated 12-O-tetradecanoylphorbol-13-acetate and phytohemagglutinin. 35S-IFN-gamma, immunoprecipitated from the crude culture fluid a monoclonal antibody, resolved by sodium dodecyl sulfate-polyacrylamide gel electrophoresis into three monomeric forms molecular weights of 25,000, 20,000, 15,500, which we designate IFN-gamma I, II, III, respectively. I most, III least, abundant both 35S-IFN-gamma chromatographically purified preparations. Changes size after glycosidase treatment suggested that contains more carbohydrate than may not be glycosylated at all. Hence, differences contents are likely to major cause heterogeneity III.
