摘要: Tetanus neurotoxin-insensitive vesicle-associated membrane protein (TI-VAMP) is a vesicular soluble N-ethyl maleimide-sensitive fusion attachment receptor (SNARE) that has been implicated in neurite outgrowth. It previously reported TI-VAMP localised the somatodendritic compartment of neurons indicating role events within dendrites. Using newly produced monoclonal antibody to improves signal/noise immunodetection, we report also present subsets axon terminals adult rat brain. Four distinctive populations labelled were identified: 1) hippocampal mossy fibres dentate gyrus and CA3, 2) striatal peridendritic terminal plexuses globus pallidus (GP), substantia nigra pars reticulata (SNr), 3) central nucleus amygdala, 4) primary sensory afferents dorsal horn spinal cord. The presynaptic localisation these locations was demonstrated by co-localisation with synaptophysin. Ultrastructural studies showed labelling over synaptic vesicles fibres, whereas it tubulo-vesicular structures multivesicular bodies pyramidal cell occurred P15, so relatively late during development. In contrast, dendritic most prominent early post-natal period. Co-localisation markers neurotransmitters TI-VAMP-positive are GABAergic GP SNr glutamatergic fibre system root afferents. Most known co-localise neuropeptides. We found met-enkephalin-immunoreactivity sizeable fraction positive GP, horn, as well few terminals. function mature remains be elucidated; could participate exocytotic molecular machinery and/or particular growth properties Thus, presence may correspond high degree plasticity characterises this pathway throughout life.
