摘要: Intestinal epithelial cells are able to producesoluble mediators that initiate or amplify inflammatoryevents in the intestinal mucosa. Interleukin (IL)-8 issuggested be a cytokine playing major role during acute and chronic processes ininflammatory bowel disease (IBD). TH-2 cytokines havebeen described as down-regulating inflammatoryresponse. We analyzed effects of IL-10, IL-13, IL-4 on IL-8 secretion intestinalepithelial cells. The human colonic cell lineCaco-2 freshly isolated cellswere used. Cells weRestimulated with IL-1beta after treatment cytokines. Levels IL-8were determined by employing enzyme-linked immunosorbentassay (ELISA). Stimulation results atime-dependent secretion. addition but not activatedepithelial resulted strong decrease IL-8secretion. Maximal inhibition required TH-2cytokines added up 60 min before simultaneous stimulatory agents. present novelfindings IL-13 strongly down-regulate IL-8secretion from Amicroenvironment containing high concentrations IL-4and may alter recruitment immune cellsto enterocytes at least partly inhibiting IL-8production. This might diminish severityof inflammatory response and, thus reduce clinical activity.
