摘要: Abstract A study was made to find the optimal conditions for titration of strong acridine orange binding sites DNA in situ by an equilibrium staining method. Low concentrations dye (≈10−6 M) and equilibration time about 1 h were found necessary. Chick erythrocyte nuclei used as a model system compare results this method with those conventional staining. Before staining, subjected acid extraction denaturation or biological activation via cell hybridization. Qualitatively similar obtained two methods, but circumvents problems dye-to-dye aggregation differences diffusion conditions, thus gives more easily interpretable data true quantitative information properties chromatin situ.
