摘要: Abstract A method for the quantitative determination of single-stranded sections in DNA was developed. Fractions denatured can be estimated using a suitable calibration curve which is obtained on basis that acridine orange bound to emits weaker fluorescence at 530 nm and stronger one 640 than does dye native DNA. The most conditions, where both types with same efficiency, were determined an ionic strength 0.01 phosphate ratio 7.5. Under these intensity added mixture heat-denatured decreased linearly as fraction increased. In relation making curve, conformation calf thymus studied. When heated chilled, only few percent total nucleotides reformed double helical structure spite considerable decrease hyperchromicity.
