Expression of antimicrobial peptides from Hyas araneus haemocytes following bacterial challenge in vitro

摘要: Circulating haemocytes play major roles in the host defense reactions of decapods, including synthesis and release antimicrobial peptides (AMPs). Unlike AMPs from insects, those decapods are constitutively expressed. This study aims to establish primary cell cultures three main haemocyte types Hyas araneus haemocytes, measure vitro expression AMP genes cells following microbial challenge. The populations were separated on Percoll gradients cultured modified L-15 medium. Expression analysis by real-time RT-PCR showed that granular producers crustin, hyastatin arasin 1 AMPs, but hyaline semigranular also show some these genes. Incubating with Aerococcus viridans var. homari (a Gram-positive bacterium) or Listonella anguillarum Gram-negative pathogen) provoked no dramatic changes gene any AMP, although there was a small (single doubling) significant increase crustin 24 h after exposure L. anguillarum, it is unclear if this biologically relevant under conditions. results presented accordance several vivo studies.
