摘要: Inflammation can contribute to brain injury, such as that resulting from ischemia or trauma. The authors have previously shown the cytokine interferon-beta (IFN-beta) affords protection against ischemic which was associated with a diminished infiltration of neutrophils and reduction in blood-brain barrier (BBB) disruption. goal current study directly assess effects IFN-beta on neutrophil infiltration, use an vivo assay relevance injury. Intrastriatal injection recombinant rat cytokine-induced chemoattractant-1, member interleukin-8 family (1 microg 1 microl), triggered massive extensive BBB disruption 6 hours later, measured using immunofluorescence microscopy magnetic resonance imaging rat, respectively. Depleting animals before prevented Treatment (5 x 106 U/kg) almost completely attenuated damage. Gelatinase zymography showed matrix metalloproteinase-9 expression ipsilateral striatum after injection. Both depletion treatment downregulated metalloproteinase-9. has already been approved for human chronic inflammatory disorder multiple sclerosis. potential value attenuate acute inflammation is considered.
