摘要: Little is known of the different freezing and thawing techniques for post-thaw survival spermatozoa in Sambar deer. So, this study determined effect seminal plasma, egg yolk glycerol extenders their concentrations, plus cooling, freezing, protocols on quality semen. Semen samples were collected by electro-ejaculation from four Thai deer stags (Cervus unicolor equinus). As evaluated progressive motility acrosome integrity removal plasma was beneficial; Tris-egg most efficient extender; a 20% concentration better than 0%, 10%, or 30%; 3% 5%, 7%, 9%. Using optimum dilution techniques, semen loaded 0.5 ml plastic straws. Cooling times ambient temperature to 5°C 3 hr resulted higher 1, 2, 4 hr. Suspending straws 4 cm above surface 15 min before plunging into liquid nitrogen suspending at 2 6 cm. For frozen semen, an intermediate (50°C, 8 sec) protocol more effective slower (37°C, 10 sec) faster (70°C, 5 sec) rates. Timed insemination following estrus synchronization 10 hinds six confirmed pregnancies 60 days. Five delivered live fawns. This provides approach cryopreservation artificial (AI), which should be valuable scientists genetics reproductive management developing countries.
