Baclofen Modulates the Expression and Release of Neurotrophins in Schwann-Like Adipose Stem Cells

摘要: Neurotrophins are a group of polypeptides that specifically influence neuronal activity during development and adult life in the central peripheral nervous system (PNS). In particular, Schwann cells (SC) PNS exert neurotrophic role following up-regulation several growth factors, including nerve factor (NGF) brain derived (BDNF). Also SC-like from adipose tissue (dASC), which have molecular functional properties similar to SC, can produce secrete NGF BDNF. Interestingly, gamma-aminobutyric acid (GABA) its receptors been also suggested as modulators myelination PNS. Therefore, it was interesting investigate whether stimulation GABA-B receptor may regulate expression neurotrophins SC dASC. Our findings demonstrated specific agonist baclofen influences secretion 2 24 h exposure lead increased both dASC, measured by western blot. Moreover, enzyme-linked immunosorbent assay showed levels released were modified after treatments. The possibility modulate potential stem cell, acting on GABAergic receptors, could represent novel pharmacological approach improve regeneration.
