摘要: MHox is a mesoderm-specific homeodomain protein that binds an A/T-rich element essential for activity of the muscle creatine kinase (MCK) enhancer. The binding site also ubiquitous Oct-1 as well myocyte enhancer-binding factor-2 (MEF2), which belongs to MADS superfamily transactivators. To determine these proteins activates MCK transcription through A/T element, we mutated this sequence such it would selectively bind MHox, MEF2, or and tested activities mutant enhancers in skeletal cells. These revealed only MEF2 able activate enhancer element. convergence on provides mechanism single DNA can mediate positive negative regulation gene reminiscent roles two classes factors control other cell-specific genes.
