New approach in RNA quantification using arginine-affinity chromatography: potential application in eukaryotic and chemically synthesized RNA

摘要: The knowledge of RNA’s role in biological systems and the recent recognition its potential use as a reliable biotherapeutic tool increase demand for development validation analytical methods accurate analysis RNA. Affinity chromatography is unique technique because versatility applications reliant on affinity ligand used. Recently, an arginine-based matrix has been effectively applied purification RNA specific mechanism molecules. This interaction suggested to be due length arginine side chain ability produce good hydrogen bonding geometries, which promote multi-contact with backbone or bases, based folding. Thus, this work presents method ultraviolet detection quantification using amino acid immobilized ligand. was validated according International European legislation bioanalytical methods. results revealed that proposed suitable detection, separation, RNA, showing precise concentrations up 200 ng/μL Furthermore, methodology demonstrated by applicability from different eukaryotic cells crude samples chemically synthesized Therefore, demonstrates multipurpose molecular biology RNA-based therapeutics.
