Azidothymidine inhibits cell growth and telomerase activity and induces DNA damage in human esophageal cancer

摘要: Esophageal cancer is one of the most common type malignancies. Telomerase activity, which absent or weakly detected in majority human somatic cells, elevated esophageal cancer. Although azidothymidine (AZT), a reverse transcriptase inhibitor, has been utilized as treatment for tumors, its role treating not confirmed. The aim present study was to determine effect AZT on telomerase activity and proliferation cell line TE‑11. A telomeric repeat amplification assay detect following TE‑11 cells with AZT. cycle distribution determined by flow cytometry. Cellular DNA damage evaluated comet an MTT demonstrated that significantly inhibited viability time‑and dose‑dependent manner. In addition, treated various concentrations exhibited significant reduction percentage G1/G0 phase, increase S phase. High doses caused damage, enhanced expression levels γ‑H2A histone family member X phosphorylated checkpoint kinase 2 cells. These results effectively inhibits vitro. growth inhibitory effects were associated G2/M phase arrest, suggesting may be clinic
