摘要: This study reports the pharmacokinetics of hydromorphone after intravenous and intramuscular administration to rhesus macaques (Macaca mulatta). Hydromorphone (0.075 mg/kg) was administered intravenously as a bolus or intramuscularly on separate occasions healthy, socially housed, reared, adult, intact male (n = 4). Blood samples were collected prior until 10 h administration. Serum concentrations analyzed with liquid chromatography-mass spectrometry. Compartment models fit time-concentration data. A 3-compartment model input in elimination from central compartment best data, whereas 1-comparment After administration, median clearance terminal half-life 37.7 (range, 33.7 47.1) mL/kg/min 142 131 218) min, respectively. The (range) 81.5 (77.2 92.5) min. Median bioavailability 92% 75% 104%). Rhesus maintained greater than equal 4.0 ng/mL for at least 2 disposition characterized by large volume distribution moderate clearance. Intramuscular resulted rapid almost complete drug absorption. Whole-body pruritus, sedation, decreased appetite observed all initial
