摘要: We previously showed substantial differences in Pseudomonas aeruginosa exopolysaccharide production vitro at oxygen tensions reflective of the right versus left cardiac circuits vivo (40 80 mm Hg, respectively; A. S. Bayer, T. O'Brien, D. C. Norman, and Nast, J. Antimicrob. Chemother. 23:21-35, 1989). However, those studies did not specifically confirm this to be characteristic P. mucoid alginate seen patients with cystic fibrosis. With a murine monoclonal antibody prepared against alginate, strongly positive immunofluorescence (IF) staining nonmucoid strain (PA-96) was after its exposure (pO2) approximately Hg; intensity IF under these conditions similar that observed phenotypically (C1712M) from fibrosis patient. In contrast, same (PA-96), pO2 40 little for alginate. Following enzyme treatment alginase, PA-96 cells exposed higher exhibiting enhanced production, as determined by staining, now no staining. Moreover, oxygen-up-regulated alginase released amounts unsaturated breakdown products (uronic acids) quantitatively typically strains treated enzyme.
